Audience
Audience holds the records of the people you email and the two ways you group them.
The section opens on a page of cards, one per screen underneath it, and each card carries one live number. That number is the only thing this page tells you that the admin's own sidebar does not. A card whose number could not be worked out says so rather than showing a zero it never measured.

Profiles
A profile is a record about a person. It carries no promise that you can email them — a healthy store holds plenty of profiles it cannot email, and that is normal rather than a problem to fix.
The card's number is the profiles that are confirmed and subscribed: ones something in your store confirmed who have also said yes to marketing email. It is not a total, and it is an upper limit on your reach rather than an exact one, for the reasons the home screen sets out.
Segments
A segment is a rule. You write the rule; who is in it follows from the rule and stays current on its own. Nobody is put into a segment by hand and nobody has to be taken out.
The number counts the segments you have, leaving out any you archived.
Lists
A list is built by placements. Someone joins because a form put them there or because you put them there, and they stay until they are taken out.
That is the difference the product keeps, and it is worth holding on to: a segment is defined by a rule and its membership is computed; a list is built by placements and members stay until you remove them.

What this section does not decide
Consent. Whether someone may be emailed is a state on their own profile, and it is the same answer whichever segment or list they turn up in. A segment is a question you ask of your profiles, never a permission: putting somebody in one gives you no right to email them, and taking them out of one takes none away.
